I participate in joint project :http://www.labflytrap.com/forum/viewtopic.php?f=42&t=20

Here I will post everything what i will do.

I want to investigate first 3 basic things:

1.Best medium for growth
2.Best concentration of hormones for multiplification
3.Best concentration of homones for rooting

4.Other things what can be researched when we know 3 above- best surcose concentration, pH, agar concentration, additions od activated charocal, antioxidants such cytric/ascorbic acid/cysteine, and mayby some other.

2 days ago i recived pack with roridula plantlets.
The same day i finished making medias, what i want to try. MS medium is most universal medium, so i want to find best concentration for further tests, and mayby when i will have more plantlets i will try also B5, WPM, others.

All medias- full MS vitamins, 30g surcose/l, 6g agar/l pH 5,6 1ml PPM/l
1. 16,6% MS
2. 25% MS
3. 33% MS
4. 50% MS
5. 66% MS
6. 75% MS
7. Knudsen C[oryginal formula with addition of EDTA to chelate iron, without vitamins.]

Yesterday i passaged plants on new medias- 5 jars with every medium, 1 roridula plant-1 jar.

They was mixed on my growrack to eliminate eventually position effect.

First observations:

-0% contamiation in the jars.
-16% MS induced offshooting with 2 plants, growth tip does'nt look well.
-Healthiest plants seems to be on 50-75% MS
-Knudson C also looks quite good

Offshooting appeard in more plants- proubably cytokinines from past work.

By now- there are some healthiest and less healthy plants in each test.

Roridula seems to accept big variety of medium concentration. At this moment- my "favorite" is 33% MS, but growth is well also at other medias...

Soon i will make a summing up with photos- now i have to make medias for next attempts.

Now i have ~35 plants of roridula [2-3 of plants from highest and lowest MS- concentrations died]

Best medium is 33-50%MS, but roridula accepts each concentration.

I will make test on different concentrations of kinetin, 6-BAP, TDZ IBA, IAA NAA, single hormone- 5-6 concentratios.Next i will make test with 2 hormones in medium. On each concentration i will use single plant from lates ster, cutted into 3-4 nodes fragments.

Finally i found some time...

I will test now only some simply methods, due to reliability when test is run on low number of jars.


Concentrations of hormones- 0.2 0.5 1.0 mg/l
Hormones- NAA IBA Kin BAP

3 jars for each concentration of single hormone. In each jar single plant cutted into 3 parts.

It seems that they like kinetin ;) They are not so hard in TC, but i think that most important will be strong root system before acclymatisation...
