Hello

I have some D.regia in in-vitro culture. I tried propagation from leaf explants on medium 40%MS with 0.015mg kinetin 0.001 IBA/l, but it falied- i didn't get any plantlets. I have to use more kinetin, yes? Now i'm trying with whole plants on 0,5mg kinetin/l,.

If anybody know hormones concentrartion for micropropagation of D.regia from leaves i will be very thankful ;)

Hi.
D. regia responds on 2mg/l 2iP + 0,1 mg/l 24D.
Medium 1/3 MS with addition 100mg/l KCl.

Yeah :D Great that it is possible. I have some 4-CPA, not 2,4-D, i also don't have 2-iP. But 2-ip have low- activity and adenine-based structure, i will try 0,5 mg 6-BAP/l + 0,1mg 4-CPA. Mayby it will work if not i have to order 2iP ;)

No ;) But BAP is also based on adenine ring,

And structure of iP is very similar, becouse there is isopenthyl group in the place of benzyl group, so their mechanism of action is similar- i hope that BA will work too.

4.02 i planted some D.regia leaves on medium with BAP and 4-CPA. Todat i saw some strike! Leaves are completly red in 90%, and doesn't look well, but in 1/10 i can see ~10 small green plantlets. Yeah ;)

Sativ, have you some news from your experience?

Since 1 month, i have 2 leaves of regia on MS/3 + 1mg/l BAP and they are green and look good. But also no signe of bud... :?

I'll wait and we'll see
