Cultivation of plants in different medias.
Become a part of a project.
I will send in-vitro plant materials for experimentation.
Necessary condition:
Publication of results of the experiment in the forum, including photographs.

A
1 / 3 MS medium+ full vitamins, 1 mg / l KINETIN, 0.2 mg / l IBA, 30g / l sugar, pH 5.65
B
1 / 4 MS medium+ full vitamins, 0.2 mg / l KINETIN, 0 , 2 mg / l IBA,30g / l sugar, pH 5.65
C
1/8 MS medium, 100mg/l myo-Inositol, 0.4mg/l Thiamine•HCI
1 mg/l 6-ã,ã-Dimethylallylaminopurine (2IP), 0,2mg/l IBA , 80 mg/l Adenine Hemisulfate, sugar 25g / l, pH 5.65

Tissue has been plated on the following media:
10% MS with full MS vitamins, 30g/L sugar, pH 5.6, 1ml/L PPM
20% MS with full MS vitamins, 30g/L sugar, pH 5.6, 1ml/L PPM
30% MS with full MS vitamins, 30g/L sugar, pH 5.6, 1ml/L PPM
40% MS with full MS vitamins, 30g/L sugar, pH 5.6, 1ml/L PPM

I focused more on the 10% and 20% MS because I have spoken to a few people that indicated that they never use more than 20% MS.

I was initially intending on using activated charcoal as well, but this will be done in the next round. I also hope to experiment with vitamin concentrations in the future as well as Knudson C media at varying concentrations.

100% contamination after only 5 days. :cry:

I'm not sure what the source was, but I think it came in transit. I've never seen this form of contamination in any of my cultures before. It appears to be some sort of pink colored bacteria.

The contamination was so bad that I didn't even attempt to save the tissue. Any chance that you have more tissue Michal?

I received the replacement tissue today. Hopefully it didn't get contaminated en route this time!

I was able to put the tissue onto the following media:
3 cultures - 50% Knudson C (original forumla) + full MS vitamins, 30g/L sugar
1 culture - 50% WPM + 50% vitamins, 30g/L sugar
2 cultures - 10% MS + full MS vitamins, 30g/L sugar
3 cultures - 20% + full MS vitamins, 30g/L sugar
3 cultures - 30% + full MS vitamins, 30g/L sugar
2 cultures - 40% + full MS vitamins, 30g/L sugar
1 culture - 50% + full MS vitamins, 30g/L sugar, 1ml/L PPM, 1.5g/L activated charcoal
pH = 5.6-5.7 for all cultures

Some of the cultures have only a bit of tissue in them, but there should be enough for them to grow. I'll post again when there's something new to report.
