Cultivation of plants in different medias. Become a part of a project. I will send in-vitro plant materials for experimentation. Necessary condition: Publication of results of the experiment in the forum, including photographs.
Medium
All medias- full MS vitamins, 30g surcose/l, 6g agar/l pH 5,6 step 1 20% MS with full MS vitamins, 30g/L sugar, pH 5.6, 6g/l agar 30% MS with full MS vitamins, 30g/L sugar, pH 5.6, 6g/lagar 40% MS with full MS vitamins, 30g/L sugar, pH 5.6, 6g/l agar step 2 Best concentration of hormones for multiplification step 3 Best concentration of homones for rooting

I received the tissue today and I was able to put the tissue onto the following media: 1 culture - 50% Knudson C (original forumla) + full MS vitamins, 30g/L sugar 1 culture - 50% WPM + 50% vitamins, 30g/L sugar 2 cultures - 10% MS + full MS vitamins, 30g/L sugar 2 cultures - 20% + full MS vitamins, 30g/L sugar 3 cultures - 30% + full MS vitamins, 30g/L sugar 4 cultures - 40% + full MS vitamins, 30g/L sugar 3 cultures - 50% + full MS vitamins, 30g/L sugar, 1ml/L PPM, 1.5g/L activated charcoal pH = 5.6-5.7 for all cultures
I'll post again when there's something to report!

A long overdue update. The H. minor tissue seemed to grow well on nearly everything I put it on. It did best on 30% to 50% MS and 50% WPM. It didn't do very well on 10% MS, 20% MS and 50% Knudson C, growing slowly and with poor color. Interestingly, the tissue didn't grow roots on any of the MS media or the Knudson C media, but it did grow roots on the WPM. Recently, I've put quite a bit of the tissue onto 10% WPM, 25% WPM and 40% WPM to verify that grows well and grows roots and I have confirmed this. Here are a few photos of the cultures before their recent replate. Attachment:

Hi flytrapcare, please have you tried 50% Knudson C with MS micronutrients or original Knudson C micronutrients ?

Yes, such results i have expected.
WPM is good rooting medium probably due higher potassium content and low salts content
Knudson C, diluted MS ( 10,20% ) are very poor mediums. Knudson C is poor in micronutrients, diluted MS in both macro and micronutrients. It will be better to use Knudson C with for example 1/2MS micro and full MS vitamins and diluted MS ( 10%,20% ) also with 1/2 MS micro and full MS vitamins.
BTW. The Knudson C media you are using is not original formula, this is later Morel modification. And you have written link to the MS vitamins not to the MS micronutrients.

Milos Sula wrote: And you have written link to the MS vitamins not to the MS micronutrients. I'm going to show my lack of knowledge here. I thought vitamins were the same thing as micronutrients In any case, I realize now that they are different. The MS mixture I'm using has macro and micronutrients and vitamins: http://www.phytotechlab.com/MSDS/M519msds.pdfBut when I cut it to 50%, I bring the vitamins back up to 100% with this: http://www.phytotechlab.com/TechInfo/M533-Info.pdfI don't even know where to purchase MS micronutrients.

For example here are micronutrients: http://www.phytotechlab.com/detail.aspx?ID=456For testing/optimalization of media is better to buy macronutrients http://www.phytotechlab.com/detail.aspx?ID=441 ,micronutrient and vitamins separately. It is good idea to test different media macronutrients first with the same vitamins and micronutrients. For example: Knudson C macro + 1/2MS micro + MS vitamins 20%MS macro + 1/2MS micro + MS vitamins WPM macro + 1/2MS micro + MS vitamins etc... Then you will select the best medium ( here i have selected 20%MS ) for your plant and you can continue testing for different micronutrients: 20%MS macro + 1/2MS micro + MS vitamins 20%MS macro + MS micro + MS vitamins 20%MS macro + 1/2WPM micro + MS vitamins 20%MS macro + WPM micro + MS vitamins after this you will select the best micronutrients for your plant and the testing continues with different vitamins...
